How this is calculated
Concentration is the vial's milligrams divided by the millilitres of bacteriostatic water you added. Volume is the dose divided by that concentration. Units are the volume multiplied by the markings per millilitre on your syringe, which is 100 for a standard U-100 insulin syringe and 40 for a U-40.
Adding more water does not weaken the dose. The vial holds the same total milligrams either way. More water lowers the concentration, so you draw a larger volume for the same amount of peptide.
