What reconstitution actually does

Melanotan II is sold as a dried powder in a sealed vial. Powder cannot be measured in fractions with any accuracy, so it is dissolved in a liquid first. Bacteriostatic water is the liquid normally used because it contains a preservative intended to limit microbial growth in a vial that will be entered more than once.

Adding water does not change how much peptide is in the vial. A 10 mg vial holds 10 mg whether you add a little water or a lot. What changes is concentration, which is milligrams per millilitre, and concentration is what determines how far up the syringe barrel a given dose sits.

That is the whole of the arithmetic on this page. Milligrams in the vial, divided by millilitres of water added, equals milligrams per millilitre. Everything else follows from that one division.

Reading the concentration table

The table above covers the single vial size commonly sold, 10 mg, against the three water volumes people use. Three things in it are worth pulling out.

First, the relationship is inverse and exact. Tripling the water from 1 mL to 3 mL cuts the concentration to a third, from 10 mg/mL to 3.33 mg/mL. There is no threshold and no rounding built into the peptide itself. The 3.33 figure is just 10 divided by 3.

Second, more water is not a weaker vial. All three rows describe the same 10 mg. A more dilute mix simply means a larger volume of liquid carries the same amount of peptide, which spreads any given dose across more syringe units.

Third, the choice of water volume is a measurement decision, not a dosing decision. Diluting further makes small amounts easier to read on a syringe, because the dose occupies more units. Diluting less means fewer injections' worth of liquid to store. This site does not tell you which trade-off to make.

Turning concentration into syringe units

A U-100 insulin syringe is marked in units, where 100 units is 1 mL. So one unit is one hundredth of a millilitre. Insulin syringes are used for peptides because the volumes involved are far too small to read on a standard syringe.

To convert, divide the dose in milligrams by the concentration in milligrams per millilitre. That gives millilitres. Multiply by 100 to get units on a U-100 syringe. The listings for Melanotan II show doses of 0.25 mg, 0.5 mg and 1 mg, and each of those reads as a different number of units depending on which row of the table you mixed to.

This is why the concentration must be recorded somewhere, on the vial or on paper, at the moment you mix. A vial that has been sitting for two weeks looks identical whether it holds 10 mg/mL or 3.33 mg/mL. The number of units for a given dose is different by a factor of three between them, and nothing about the vial reveals which one you made.
